Abstract

Breast cancer is a deadly disease which most of experienced by women. Areca nut can be used as an alternative cancer treatment. Rind of areca nut has a few compounds such as flavonoids, phenolics, alkaloids and terpenoids. This study is aimed to determine the cytotoxic effect, IC50 values, knowing selectivity index, and determine the most effective between ethanol extract and ethyl acetate fraction of areca nut rind against breast cancer T47D cell lines. The extraction process was nut rind maceration method using ethanol 96% and followed by fractionation to obtain fraction of ethyl acetate. Cytotoxic test on breast cancer cell T47D and Vero cell lines used MTT assay method and absorbance was read at ELISA reader at λ 595 nm. This study used 7 concentrations were 1000, 500, 250, 125, 62.5, 31.25, 15.63, 7.81 µg/mL. IC50 used as parameter which derived from linear regression between log concentration versus % viability and selectivity index (SI) is obtained from comparation between IC50 of Vero cell and IC50 of T47D cell. The results of the cytotoxic activity assay of ethanol extract and ethyl acetate fraction of areca nut rind had less potent cytotoxic effects against T47D cells with IC50 value of 467.3457 µg/mL and 297.7506 µg/mL. Selectivity index of ethanol extract is gotten value of 0.437 which show there were no selectivity. Based on the test results of Independent T-test, ethanol extract and ethyl acetate fraction there were no significantly differences in show cytotoxic effect.
