Abstract

Piroxicam is a non steroidal anti inflammatory drug (NSAID) commonly used in treatment of acute rheumatoid arthritis and osteoarthritis. The problem of piroxicam is poor solubility with delayed onset of action. This can be overcome by making inclusion complexes with β-cyclodextrin and create drug delivery systems capable of accelerating the disintegration and dissolution of the tablet piroxicam as Orally Disintegrating Tablets (ODT). The combined of Ac-Di-Sol® and Kollidon® CL as superdisintegrant and Avicel® PH 102 as filler binder is expected to generate ODT piroxicam with good physical properties and sweet taste. Optimization by simplex lattice design method was applied to optimize ODT piroxicam in complex inclusion with β-cyclodextrin. Ac-Di-Sol®, Kollidon® CL, and Avicel® PH102 were used as independent variable. The optimum area was determined using superimposed contour plot of hardness, friability, wetting time, disintegration time, and dissolution at 3 minute using Design Expert software. The result showed that increased of Ac-Di-Sol® and Kollidon® CL, also combination both of them as superdisintegrant will reduced wetting time, disintegration time, and increased dissolution. Increased of Avicel® PH 102 as filler binder will increased flow time, incrased in hardness, reduced friability. Combination of Ac-Di-Sol®, Kollidon® CL, and Avicel® PH 102 will reduced hardness, increased friability, increased in wetting time and disintegration time, also reduced dissolution. The optimum of ODT piroxicam with optimum physical properties and sweet taste was Ac-Di-Sol® 2,85%, Kollidon® CL 12,77%, and Avicel PH® 102 84,38 %.
