Abstract

Diabetes mellitus is a disease characterized by hyperglycemia and several metabolic disorders caused by absolute or relative deficiency of insulin and/or insulin resistance. The purpose of this study was to determine antihyperglycemic and pancreas regeneration activity of fractions from mahogany seed extract. This study used ten groups of rats and streptozotocin-nicotinamid induced 50/110 mg, except group 1 was normal control group. Group 2 was given a CMCNa 1%, group 3 was given of Glibenclamide, group 4 was given a mahogany seed extract 300 mg/kgBB, group 5-6 was given n-hexane fraction in two doses, group 7-8 was given ethyl acetate fraction in two doses, dan group 9-10 was given water fraction in two doses, was 30 and 60mg/kg. Test material was administered orally for 14 days, then seen an increase in body weight of rats, a decrease in blood glucose levels, and improvement of pancreatic histopathology profile with Hemaktosilin-eosin and immunohistochemistry staining. The results showed that ethanol extract mahogany seeds 300 mg / kg has the ability to increase body weight as well as the effect of glibenclamide, with increases glucose utilization in peripheral tissues. As for the decrease in blood glucose levels and pancreatic histopathological profile improvement is seen by the data in diameter and density of islet cells, as well as the presence of insulin in the islet cells, generated by n-hexane fraction mahogany seeds 60 mg/kg. However, based on statistical analysis, extract, fractions and positive control group, equally significant impact as antihyperglycemic and pancreas protector.
