Abstract

Yacon leaves are plants that can be used as an alternative cancer treatment. The aim of this study was to determine the cytotoxic activity of extract, n-hexane fraction, ethyl acetate fraction and water fraction yacon (Smallanthus sonchifolius) on T47D cells and to determine the effect of expression of p53 and Bcl-2 on yacon leaf active fraction (Smallanthus sonchifolius). The extract was obtained by the soxhletation method with 70% ethanol solvent. ethanol extract is then fractionated with liquid-liquid partition. Cytotoxic tests were carried out using T47D cancer cells and vero cells with the MTT assay method and read the absorbance with an ELISA reader. To determine the effect of p53 and Bcl-2 protein expression, immunocytochemical tests were performed. The results showed that the extract and ethyl acetate fraction of yakon leaf had cytotoxic activity against T47D breast cancer cells with IC50 values of 73,47 /g / mL and 56,84 /g / mL, while the water fraction and n-hexane fraction did not have cytotoxic activity with an IC50 value> 100g / mL. Yakon leaf extract is able to increase the expression of p53 and Bcl-2 protein at a concentration of 36,73 g / mL - 146,94 g / mL and the ethyl acetate fraction can increase the expression of p53 and Bcl-2 proteins at a concentration of 28,42 g / mL - 113,68 g / mL.
