Abstract

Bacteria in the oral cavity are associated with oral health as well as dental caries. Streptococcus relies on biofilms to survive and survive in the oral cavity. The main factor causing dental caries is S. mutans colonization in dental biofilms. Alamanda leaves contain flavonoid chemicals which have the potential to inhibit the intercellular adhesion genes of icaA and icaD, which are one of the factors in biofilm formation. The aim of this study was to determine the antibiofilm activity of n-hexane, ethyl acetate and water fractions from ethanol extract of alamanda leaves against S. mutans ATCC 25175. Alamanda leaf powder was extracted by maceration method using 96% ethanol solvent. The extract obtained was fractionated with n- hexane, ethyl acetate and water solvents. Extracts and fractions were tested for antibiofilm activity against S. mutans ATCC 25175 by the Microtiter Plate Biofilm Assay method using a microtitterplater flat-bottom polystyrene 96 wells followed by a microplate reader at λ = 550 nm. The concentrations used were 2, 4, 6, and 8 mg/mL. Statistical analysis was performed using the ANOVA test and continued with the posthoc test. The results showed the extract and fraction of ethanol extract of Alamanda leaves could inhibit the formation of biofilm S. mutans ATCC 25175. IC50 values of the n-hexane, ethyl acetate and water fractions from the extract of alamanda leaf were 1.69; 1.38; 1.65; and 1.90 mg/mL. The greatest inhibitory activity of S. mutans biofilms is shown by the ethyl acetate fraction which is also the most active fraction with an IC50 value of 1.38 mg/mL.
