Abstract

Metoclopramide HCl is an anti-emetic drug that is classified as a dopamine receptor antagonist. Drugs with rapid onset are needed to treat nausea and vomiting, so metoclopramide HCl is suitable to be made in the form of FDT dosage form because it breaks down quickly in the mouth. This study aims to obtain the optimum FDT formula of metoclopramide HCl using superdisintegrant sodium starch glycolate and filler-binder ludipress®. This study used the simplex lattice design method with 2 factors, namely SSG as a superdisintegrant and ludipress® as a filler-binder. The formula is made 3 variations in the sequence concentration 0%: 100%; 50%: 50%; 100%: 0%. The critical point parameters and optimum formula are obtained from the response of hardness, friability, disintegration time, Q3, and DE3 using one sample t-test statistical test. The results showed that variations in the concentration of 3 metoclopramide HCl formulas produced good physical quality tablets. Sodium starch glycolate has an effect on increasing friability and disintegration time, while ludipress® has an effect on increasing hardness, Q3, and DE3 of tablets. The proportion of sodium starch glycolate 2% (3 mg) and ludipress® 58% (87 mg) produce the optimum formula of FDT metoclopramide HCl with the best physical tablet quality.
