Abstract

UV-A radiation induct disorder to the skin called photoaging. Photoaging need to treatment with antioxidant agent. SOD is the first linear antioxidant in the treatment of free radicals. This study aimed to find out anti photoaging activities of SOD extract of corn (Zea mays L.) kernels on radiated UV-A rays 3T3 fibroblast cells. SOD extraction of corn kernels was done by the centrifugation method, precipitation and dialysis. SOD extract of corn kernels was measured in total protein content through the Bradford method, followed by measurement of SOD activity by the WST-1 method. Anti photoaging activity SOD extract of corn kernels was done on radiated UV-A rays 3T3 fibroblast cells with parameters were cell viability and collagen deposition. Anti photoaging activity tested use cell control, negative control sampel and positive control. The results of anti photoaging activity measurement of SOD extracts of corn kernels were analyzed by One Way ANOVA by Tukey's difference significance test. Total protein content showed that highest value was SOD extract of corn kernels in precipitation of ammonium sulphate with a concentration was 40-60%. The result of SOD activity showed that the highest percent inhibition value was SOD extract of corn kernels in precipitation of ammonium sulphate with concentration was 60-90%. The result of anti photoaging activity SOD extract of corn kernels showed significant difference in the value of percent cell viability and percent collagen in fibroblast cells between the extract group and the negative control group with the highest percent cell viability and percent collagen value in SOD extract corn kernels with the concentration was 100 mg/ml. EC90 value for cell viability and collagen deposition parameters respectively 12,57 mg/ml and 69,47 mg/ml. Key words: Corn kernel, Anti photoaging, Collagen, SOD, 3T3 Fibroblast cell
