Abstract

Dental caries is a disease that is often found in the oral cavity. The main agent that causes dental caries is Streptococcus mutans. Pucuk merah leaves (Syzygium myrtifolium Walp.) contain compounds which have antibacterial activity, the main compounds in pucuk merah leaves are flavonoids, alkaloids, steroids, terpenoids, saponins, and phenols. The purpose of this study was to determine the antibacterial activity of extract and essential oil of young and old pucuk merah leaves against Streptococcus mutans ATCC 25175. The extract was obtained by maceration using 96% ethanol solvent, and isolation of essential oils was carried out by steam-distillation. Antibacterial activity testing uses diffusion and dilution methods. Screening antibacterial activity of extracts and essential oil of young and old pucuk merah leaves through diffusion method by measuring the diameter of the inhibitory zone. The most active test sample is continued with dilution method to determine KHM and KBM. The diameter of the inhibition zone was analyzed by one way ANOVA method. The results showed that young and old leaves of extracts and essential oils had antibacterial activity. The diffusion method on essential oils of young leaves concentration of 60% had the greatest antibacterial activity against S. mutans ATCC 25175 with inhibitory zone diameter of 17.40 mm. The dilution method in essential oils of young leaves has a KBM value of 60%. Keywords : pucuk merah, Streptococcus mutans, diffusion, dilution
