Abstract

Endophytic bacteria is symbiotic microorganism that live in plant tissues and doesn’t cause negative effects on its host plant. Endophytic bacteria is an alternative producer of bioactive compounds that have the potential as an antibacterial. Therefore, this study aims to determine the antibacterial activity of Pseudomonas knackmussii and Bacillus siamensis endophytic bacteria in inhibiting Staphylococcus aureus ATCC 25923 and knowing the optimum time fermentation of endophytic bacteria which had the greatest antibacterial activity. This research was conducted in several stages, they were identification of pathogenic bacteria and endophytic bacteria, fermentation of endophytic bacteria to produce bioactive compounds and antibacterial activity tests using the disc diffusion method. Several treatment groups in the study were ciprofloxacin 5 µg as positive control, paper disc without antibacterial compounds as negative control and supernatant from endophytic bacterial fermentation for 4 days as a test group. Data was analyzed using One Way ANOVA method. The results show that endophytic bacteria from taro tubers has antibacterial activity against S. aureus ATCC 25923. The optimum time fermentation of endofit bacteria had the greatest inhibition activity reached by the third day. Fermentation of endophytic bacteria on the third day of P. knackmussii with zone inhibition is 9,69 mm and B. siamensis is 9,20mm.
