Abstract

Treatment of thalassemia in the form of blood transfusions causes a buildup of iron iron overload (OI) in the organ and cell damage. The administration of deferoxamine as iron kelator causes many side effects and is expensive. Therefore, treatment with safer and more affordable materials is needed by utilizing natural ingredients that have iron chelating effects. The purpose of this study was to determine the activity of ethanol extract of kweni mango leaves as iron chelating agents in ferro sulfate (FeSO4) induced rats by looking at SGPT levels, measuring ferritin levels, and seeing liver histopathology. This study invitro and invivo consisted of 6 groups, each group consisting of 5 male white rats Wistar strain aged 2-3 months with a body weight of 150-250 g. Group 1 normal group (male white mouse given saline 50 mg / kg BB), group 2 negative control (male white mouse given ferro sulfate 200 mg / 200g BB), group 3 positive control (male white mouse given ferro sulfate 200 mg / 200g BB), group 4 extract dose 40 mg / 200 g BB (male white rats given ferro sulfate 200 mg / 200g BB and ethanol extract dose 40 mg / 200 g BB), group 5 extract dose 80 mg / 200 g BB (male white rats given ferro sulfate 200 mg / 200 g BB and extract dose 80 mg / 200 g BB), group 6 extract dose 160 mg / 200 g BB (male white rats were given ferro sulfate 200 mg / 200g BB and extract dose 160 mg / 200 g BB). Treatment for 28 days, then carried out iron chelating test (Ferrous Ion Chelating), measurement of SGPT activity, measurement of ferritin levels in the Elisa method, and liver histopathology. The results were analyzed statistically using the Shapiro Wilk normality test then one way ANOVA analysis to test the mean (average) differences in data from more than two groups. The results showed that the ethanol extract of kweni mango leaves contained alkaloid compounds, flavonoids, steroids, tannins and mangiferin. Ethanol extract has an activity as an iron chelating agent invitro which is obtained IC ₅ₒ of 20.81 μg / ml. Invivo activity testing at a dose of 160 mg / 200 g BB provides the greatest activity in reducing ferritin levels, decreases SGPT activity and is most powerful in inhibiting liver cell damage.
