Abstract

Water spinach root is widely used as antibacterial because of the phenolic compounds contained in it. Soursop leaves (Annona muricata L.) can inhibit bacterial growth with the content of active compounds such as alkaloids, tannins, flavonoids. This study aimed to determine the effect of the combination of ethyl acetate fraction of water spinach root and soursop leaf in toothpaste preparations on physical properties, stability and antibacterial activity against S. mutans ATCC 35668 bacteria and testing of acrylic teeth. Organoleptic tests such as smell, taste and color descriptively. Physical properties with dispersion, viscosity, foam height and pH parameters were analyzed using Shapiro Wilk. Physical stability was tested with the same parameters for one month analyzed using Shapiro Wilk. The zone of inhibition of fractions and toothpaste preparations were tested using One Way Anova. Ethyl acetate fraction of water spinach root and soursop leaf with a concentration of 10%, tested by diffusion using wells, negative control used DMSO 2% while positive control triclosan 5%. The inhibition zone of ethyl acetate fraction of water spinach root is 7.88 mm, while soursop leaves are 7.66 mm. Formula 1, 2 and 3 each produce inhibitory zones of 14.10; 14.77; and 13.66 mm. The antibacterial activity test on acrylic teeth showed that the formula 2 killed the most S. mutans ATCC 35668 bacteria. The three formulas in the test were stable physical properties at viscosity, foam height and pH except dispersion. and physical stability of the formula is not stable on all parameters during storage of room temperature, oven temperature and refrigerator temperature except pH parameters.
