Abstract

Dexamethasone is one of a drug with a solubility practically insoluble in water. Tween 80 can make lower interfacial tension between drug and medium, once to form micelles so as drug molecule will be carried by micelles dissolve into the medium (Martin et. al 1993). Wicaksono (2006) revealed that PVP K30 as a hydrophilic binder as so can to increase the dissolution. This study aims to determine the propotion of tween 80 and PVP K30 to obtain optimum formula with good physical quality tablet and rapid dissolution using Simplex Lattice Design method. Dexamethasone tablet is made by wet granulation method. Based on the Simplex Lattice Design method, it is made by using three formulas. The proportion of formulas using Tween 80 and PVP K30 are F1(100%:0%); F2(50%:50%); F3(0%:100%). Each formula is made granule and tablet, then tested against to critical point parameters which are optimized using Design Expert 8.0.6. program, including flow rate, hardness, friability, disintegration time and dissolution. Optimum formula then validated by comparing the response of a critical point between theoretical and experimental result using t-test 95% significance level. Result shows no difference between prediction and experiment with Simplex Lattice Design method. It gains the optimum formula for 41,5% Tween 80 and PVP K30 at 54,9%. Tablets which are produced to meet the quality requirement of physical and has a faster dissolution rate compared with generic tablet on market.
