Abstract

Captopril is an angiotensin converting enzyme inhibitor as hypertensive treatment with half-life about 2 hours. Captopril is most stable at acidic condition and unstable in intestine or pH increases with degradation reaction. These indicate a promising potential as gastroretentive drug delivery system to improve the bioavailability and prolong the drug release. This research purposed to optimize and evaluate the influence of xanthan gum, sodium bicarbonate-citric acid as effervescent components and hardness on floatation behavior and drug release of gastro-retentive captopril tablet with floating system. A 23 full factorial design (8 runs) was applied to optimize the floating captopril tablet using xanthan gum, sodium bicarbonate-citric acid, and hardness as independent variables. Optimum area was determined by superimposed contour plot of floating lag time, cumulative drug release at 60 minutes, and drug release rate using software Design Expert®. Hardness was the most dominant factor affected the floatation behavior and drug release. Enhancement the xanthan gum and hardness level reduced the initial burst release effect and drug release rate. Enhancement of citric acid increased the drug release at initial time and enhancement of sodium bicarbonate increased amount of the drug released. Based on superimposed contour plot, the optimum area was in range of xanthan gum 58 – 100 mg, sodium bicarbonate 45 – 63 mg, citric acid 7 – 25 mg and hardness at 7,0 – 9,8 kg. Keywords : xanthan gum, effervescent components, hardness, captopril floating tablet
