Abstract

This study used natural ingredients black cumin seeds (Nigella sativa). Black cumin has activity as antidiabetic, anticancer, imunomodularotor and hepatoprotective. Black cumin extract was reported had antioxidant activity with IC50 values of 168.8 ± 2.1 μg/mL. The purpose of this study was to determine the antioxidant activity of extracts, fractions and subfractions of black cumin seeds and identification of chemical compounds in KLT group with various reagents. Black cumin seed extract obtained by maceration and fractionated with the solvent n-hexane, ethyl acetate and water. Fraction with the highest antioxidant activity sparated by vacuum liquid chromatography. Extracts, fraction and subfraction tested the antioxidant activity by DPPH method (1,1-Diphenyl-2-picrylhydrazyl) and calculated values of antioxidant activity (IC50). Quercetin was used as a positive control. The test results showed the antioxidant activity of ethanol extract of black cumin seed has IC50 value of 621.26 ± 2.57 μg/mL. Tests on the third fraction, ethyl acetate fraction showed to have the best IC50 value is 156.49 ± 0.97 μg/mL. Fraction ethyl acetate to produce 7 subfraksi combined with the results of antioxidant activity showed subfraksi D has the best IC50 value is 42.55 ± 0.05 μg/mL. Subfraksi D black cumin seeds contain alkaloids, flavonoids, steroids, terpenes, and phenolic compounds. Key word : black cumin seed, antioxidant, DPPH, ethyl acetat subfraction
