Abstract

Simvastatin isa statin drug, working as a very effective cholesterol-lowering agent.Simvastatin is a drug belonging to BCS class II. Simvastatin has a low solubilty in water of 0,003 g/L so it was the reason to make nanoemulsion preparations with SNEDDS methods to improve drug bioavailability in human body.The aim of this research is to obtain the best formula on the combination of oil, surfactant and cosurfactant based on characterization test and physical stability ie emulsification time, drug loading, transmittance percentage, emulsification time, PSA test and cycling test and to see dissolution test result of SNEDDS simvastatin formula against pure simvastatin. This research uses seven formulas obtained based on Simplex Latice Design concept consisting of combination of olive oil, tween 80 as surfactant and PEG 400 as cosurfactant, then seven SNEDDS formula tested characterization and physical stability ie emulsification time, drug loading, percent transmittance, emulsification time , PSA test and cycling test. The best result of the next formula is dissolution test compared with pure simvastatin. The best simvastatin SNEDDS formula (F5) with olive oil content 0% (1.3 mg), tween 80 50% (5.1 mg), PEG 400 50% (3.6 mg) showed an emulsification time of 19 seconds, transmittance percent of 1.8% drug loading of 206,685 ppm, 251.0 nm particle size with a PDI value of 0.658, zeta potential -21.2 mV and clear cycling test results. The dissolution test of SNEDDS simvastatin (F5) on phosphate buffer media pH 7.0 depicts a percentile increase of 101.91% within 45 minutes higher than the pure simvastatin of 29.88%.
