Abstract

Glibenclamide is an oral antidiabetika used in the treatment of mild to moderate diabetes mellitus that has low solubility in water and has a high permeability in the intestine that is categorized in Class II Biopharmaceutical Classification System (BCS). These drugs are marketed in a variety of generic and branded preparations, allowing for different formulations and fabrication methods. This study attempted to compared between generic and branded products in physical tablets quality test and tablet dissolution test. The test product used 5 glibenclamide 5 mg tablets on the market, there were 3 branded products (ODA, ODD and ODE) and 2 generic products (OGB and OGC). Dissolution tests were performed using a type 2 with speed of rotation 50 rpm in 900 ml of HCl - methanol 0.1 N pH 1,2 with 37˚C ± 0.5 temperature. Determination of dissolution rate using UV-VIS spectrophotometer at 300 nm wavelength. The results of the Q45, DE45 and F2 parameters show that all products produce different values. Q45 without significant difference are ODA (99.14%), OGB (95.57%), OGC (82.78%), ODE (81.35%), ODD (94.56%). And obtained the value of DE45 are ODA (37.45%), OGC (34.08%), ODD (32.76%), ODE (36.52%), OGB (37.17%). And the most similar F2 values between branded and generic products is ODA with OGB of 90.85%.
