Abstract

Meloxicam is a class of NSAID drugs that are practically insoluble in water and therefore need to be increased solubility with the β-cyclodextrin inclusion complex. Meloxicam is used to treat osteoarthritis and rheumatoid arthritis require fast-acting therapy, making it suitable for fast disintegrating tablet preparations (FDT). This study aims to optimize and evaluate the effect of primellose®, pearlitol® SD200 and effervescent components in order to obtain FDT with the optimum parameters as require. Inclusion complex of meloxicam in the β- cyclodextrin was prepared using keneading method in 1:2 drug to β- cyclodextrin molar ratio. Direct compression method was applied in tablet preparation with primellose®, peralitol® SD200, and effervescent componen as the independent variable. The optimum area was determined using superimposed contour plot of combination of disintegration time, weting time, friabiliti, hardness, the amoundt of drug released at 0,5 minute and dissolution efficiency using Design Expert Software. The results showed that increased of Primellose® and effervescent components, also combination both of them as superdisintegrant will reduced wetting time, disintegration time, and increased dissolution tablets. Increased of Pearlitol® SD200 as filler binder will increased flow time, increases in hardness, and reduced fragility of meloxicam FDT. Primellose®, Pearlitol® SD200 and effervescent combinations will reduce hardness, increase friability, increase in wetting time, and disintegrate time, also reduced dissolution. The formula optimum of FDT Meloxicam with optimum physical properties and sweet taste was combination Primellose® 2,000 mg, Pearlitol® SD200 37,083 mg, and effervescent component 10,917 mg.
